Firefox with Adblock and Noscript along with some common sense will take care of things in most cases, but if you really want to be totally safe...
Install Linux on a second partition and boot into it to do anything internet-related. Keep Windows around but use it only for games like World of Warcraft. Of course, I suppose one could also install linux inside of a virtual machine (like VMWare or Virtual PC), but either way, it's going to make it nigh impossible for your Windows install to get poisoned. Also, as a side effect, if your Windows install DOES manage to get infected somehow, all the files, passwords, etc you have on the Linux side are totally safe since Windows doesn't know what the hell an ext2/ext3 (Linux) partition is.
